Elucidating the role of proteasomal ubiquitin receptor Rpn13 in growth and development
The 26S proteasome is a large macromolecular protease complex responsible for the degradation of the majority of cellular proteins. Intrinsic proteasomal ubiquitin receptors Rpn10/S5a and Rpn13/ADRM1 bind ubiquitinated cargo, as well as ubiquitin-like domains of proteasomal shuttling receptors, thus recruiting the proteins destined for degradation to the proteasome. We have previously identified Rpn13 as proteasomal ubiquitin receptor and are currently working on elucidating specific roles of this protein in the growth and development. To that aim we have generated Rpn13 knock-in and conditional knock-out mouse models that we are currently studying. I will present our current knowledge about the role of Rpn13 in mouse development and point out further directions of our project.
