Systemin/jasmonate-regulated systemic defense signaling in tomato

In plants wound-induced local and systemic expression of defense related genes are regulated by intercellular mobile signals. Wound-inducible defensive proteinase inhibitors in tomato plants pro-vide an attractive model system to identify the long-distance wound signals and to dissect the signal transduction pathway leading from injury to marker gene expression. Genetic and molecular approaches have been applied to identify novel components of the systemin/jasmonic acid-mediated wound response pathway. Using map-based cloning several genes were isolated such as Spr2 encoding an ω–3 fatty acid desaturase required for the production of linolenic acid, the metabolic precursor of jasmonic acid, or Jasmonic Acid Insensitive 1. We showed that jasmonic acid biosynthesis in wounded leaves is required for the production of a long-distance signal for PI expression, but not the recognition of this signal in responding leaves. Conversely, jasmonic acid action is necessary for recognition of the transmissible signal in responding leaves, but not for its production in wounded leaves. Our results challenge the existing paradigm that the systemin signal is the sole long-distance signal for wound-induced defense gene expression.
