Over the past two decades magnetic resonance spectroscopy (MRS) has been established as an invaluable non-invasive method in clinical research. While magnetic resonance imaging helps to identify structural changes in the brain, spectroscopic techniques enable the detection of several brain metabolites such as the excitatory neurotransmitter glutamate and the inhibitory neurotransmitter y-aminobutyric acid (GABA). However, due to their low concentration and the inherently low sensitivity of MRS, detection and quantification of theses brain metabolites in vivo is rather challenging and often requires sophisticated techniques such as spectral editing, which filters the metabolite signal of interest from a background of other resonances. In our FRIAS-funded project we apply the MEGA-PRESS editing sequence for the detection of metabolic abnormalities associated with primary insomnia. Previous studies suggest that hyperarousal plays a pivotal role for the development and maintenance of this disorder, hypothesizing an altered GABA metabolism, which we aim to detect by means of MRS experiments in insomnia patients and healthy subjects.
