Disease Tolerance as a Defence Strategy to Age

Selected through evolution, immune-driven resistance mechanisms are the prevailing host defence strategy against infectious diseases. By contrast, disease tolerance mechanisms limit disease severity by preventing tissue damage or ameliorating tissue function without interfering with pathogen load. His holds also true regarding sterile inflammation caused by autoimmune diseases or metabolic stress that characterises ageing.

Tissue damage control may underlie many of the protective effects of disease tolerance. We will explore the mechanisms of cellular adaptation that underlie tissue damage control in response to sterile inflammation, taking examples in the field of kidney and cardiovascular diseases, integrating both stress and damage responses. Finally, we will discuss the potential impact of targeting these mechanisms in the prevention of accelerated ageing.
