Interaction Graphs and Logical Networks as Modeling Frame-works for Signal Transduction Networks
I give an introduction to interaction graphs and Boolean networks, two related frameworks for qualitative modeling of signal transduction networks. Using EGFR/ErbB and HGF signaling in hepatocytes as examples, I demonstrate that these modeling concepts help to uncover important structural properties of signaling networks and that they deliver verifiable predictions on network behavior driving the iterative cycle between experiment and modeling. As one important result network topologies can be selected that are consistent with experimental data and which can then be used as a starting point for a more detailed mathematical modeling of the studied pathways.
