Protein transport in cells: from proteomics to biogenesis of powerhouses

Cells are typically divided into numerous compartments, the cell organelles. Proteins are essential for the function of each organelle. However, most proteins are synthesized in the cytosol of cells, i.e. outside the cell organelles. Cells have evolved sophisticated systems to direct newly synthesized proteins into the correct organelle. Mitochondria, the cellular powerhouses, contain about 1000 different proteins. Proteomic studies combined with biochemical and genetic analysis revealed an unexpected diversity of transport routes and mechanisms of how proteins are imported into mitochondria. After passing through a central entry gate, the precursor proteins are transported by four different machineries to the different internal compartments of mitochondria.
