Fluorescent biosensors for cell regulation pathways

Live cell health and function is controlled by interactions between thousands of types of proteins that are parts of regulatory pathways. We are developing a toolkit of fluorescent probes (or biosensors) for quantifying these regulation pathways. The biosensor development program targets membrane surface protein traffic, protease activity, protein-protein interactions, calcium levels pH and membrane potential. The new biosensors are based on combining an engineered single chain antibody (FAP: Fluorescence Activating Protein) and a fluorogen that is not fluorescent in the medium but becomes highly fluorescent when associated with the FAP. FAPs have been created with different fluorogen dissociation constants and different structures. Fluorogens have been synthesized with emission wavelengths from 450-850 nm.
